Following the route of the core section of the West Coast Main Line between London and Glasgow. It is 347 nautical miles in actuality, whereas this route is 339 nm. Since this will likely be flown VFR by most people, I've only included waypoints for the 22 stations. It shouldn't be too hard to follow the railway line precisely if you so choose.

The halfway point is around Manchester, so if you want to break up the flight over a couple of stints, there are a few airports in the vicinity at which to take a break.
